In vitro culture of Perkinsus atlanticus, a parasite of the carpet shell clam Ruditapes decussatus.
Abstract
La capacidad de cultivar in vitro grandes cantidades de parásitos facilita la investigación biológica, metabólica y morfológica que de otra manera sería difícil de llevar a cabo. Hemos establecido cultivos continuos in vitro del protista Perkinsus atlanticus de almeja fina, y además hemos estudiado variables que afectan a la proliferación del parásito como medio de cultivo, tamaño de inóculo, temperatura y salinidad. El parásito se adaptó a muy diferentes medios de cultivo, salinidad y temperatura, y la densidad del inóculo no afectó a la concentración final celular alcanzada in vitro. Describimos la morfología de P. atlanticus cultivado, y comparamos sus condiciones óptimas de crecimiento con las de P. marinus, parásito de la ostra Crassotrea virginica. Para asegurar la conservación a largo plazo del P. atlanticus cultivado optimizamos sus condiciones de criopreservación.
